**Background**
Inflammation and oxidative stress are central drivers in the progression of various pathologies, including cancer, diabetes, and neurodegenerative diseases. Cyclooxygenase-1 (COX-1) and the nuclear factor kappa-light-chain-enhancer of activated B cells (NF-κB) pathway play pivotal roles in mediating the inflammatory response. Specifically, the activation of NF-κB leads to the expression of inducible nitric oxide synthase (iNOS) and other pro-inflammatory cytokines, contributing to tissue damage and tumor proliferation. Consequently, targeting these pathways is a primary strategy for developing therapeutic interventions. In this context, we will introduce a potent bioactive flavan-3-ol – (-)-Epicatechin.
**Definition**
(-)-Epicatechin is a COX-1 inhibitor with an IC50 value of 3.2 μM. According to the (-)-Epicatechin description, it also acts by blocking the nuclear localization of the p65 subunit of NF-κB to inhibit iNOS expression.
**In Vitro and In Vivo Studies**
The (-)-Epicatechin biological activity has been extensively characterized across various cell lines and animal models. In vitro, (-)-Epicatechin exhibits >95% inhibitory activity against COX-1 at a concentration of 70 μg/mL. In RINm5F cells, it effectively blocks the inhibition of insulin release induced by IL-1β. Furthermore, it inhibits the proliferation of Jurkat T cells and Hodgkin’s lymphoma cells by preventing NF-κB from binding to DNA. In the context of (-)-Epicatechin Cancer research, combining 150, 200, or 250 μM of (-)-Epicatechin with 20 μM Panaxadiol in human colorectal cancer HCT-116 cells resulted in growth inhibition of 97%, 95%, and 95%, respectively, while increasing apoptosis levels by 11.9% to 25.8%. Other cellular effects include the inhibition of COX2 mRNA expression in DLD-1 cells (IC50 415 μM) and antiaggregatory activity in human platelets (IC50 22.22 μg/mL for collagen-induced aggregation).
Regarding (-)-Epicatechin in vivo application, animals receiving 1 mg/kg of the compound via oral gavage twice daily, combined with 15 days of treadmill exercise, showed significant improvements in physical performance. Specifically, treadmill performance increased by approximately 50% and in situ muscle fatigue resistance was enhanced by roughly 30%. In conclusion, (-)-Epicatechin is a versatile inhibitor of COX-1 and NF-κB with significant potential in treating inflammatory diseases and enhancing muscle performance.
